螺旋藻多糖在2215细胞培养中对乙型肝炎病毒表面抗原和e抗原及HBV-DNA的抑制作用
Inhibition of Spirulina Polysaccharide to HBeAg,HBsAg and HBV DNA Cultured in 2215 Cell Line
【摘要】 研究螺旋藻多糖在乙型肝炎病毒基因转染的人肝癌细胞系 2 2 15细胞培养中对细胞的毒性、对HBe Ag和 HBs Ag分泌的影响及对细胞 HBV-DNA的抑制效果。 3批试验结果均表明 :螺旋藻多糖加入细胞培养12 d对细胞的半数中毒浓度 >4 mg/ml,最大无毒浓度为 1.5± 0 .7mg/ml;抑制细胞分泌 HBe Ag 50 .4 % ;抑制HBs Ag4 2 .7% ;对 HBe Ag和 HBs Ag的半数有效剂量 IC5 0 分别为 2 .62± 0 .2 5mg/ml和 2 .90± 0 .18mg/m l,治疗指数分别为 >1.53和 >1.3 8,对 HBV-DNA亦有一定抑制。说明螺旋藻多糖在 2 2 15细胞中无毒浓度可抑制HBe Ag、HBs Ag及 HBV-DNA。
【Abstract】 Spirulina polysaccharide(sp) solution was added to 2215 cell line transfected with HBV.Twelve days later,toxic concentration 50(TC 50 )of sp to 2215 cell exceeded 4 mg/ml and maximum non toxic conentration(TC 0) reached 1 5±0 7 mg/ml.Inhibition rate of sp to HBeAg and HBsAg secretion were 50 4% and 42 7%,respectively.Median infective concentration(IC 50 )of sp to HBeAg and HBsAg secretion equaled 2 62±0 25 mg/ml and 2 90±0 18 mg/ml,respectively,with therapeutic indices more than 1 53 and 1 38.Sp also inhibited HBV DNA.All these suggested that sp at non toxic concentration could suppress HBeAg and HBsAg expression and decrease the HBV DNA level.
