水母雪莲愈伤组织的多态性及其再分化条件的研究

【摘要】 以水母雪莲 ( Saussurea medusa Maxim.)的幼叶为外植体 ,接种于 MS补加 0 .2~4 .0 mg/ L NAA、0 .2~ 4 .0 mg/ L 6-BA的不同组合的培养基上培养 3周 ,即形成大量的原始愈伤组织 ,其中以 MS+2 .0 mg/ L NAA+0 .2 mg/ L 6-BA培养基上的诱导效果最好 ,诱导率可达1 0 0 % .水母雪莲原始愈伤组织呈多态性 ,经继代培养可分离出白色水状、淡黄绿色致密的和紫红色疏松的 3种愈伤组织 ,它们在分化能力上有明显差异 ,白色和淡黄绿色的愈伤组织均无分化能力 ,紫红色愈伤组织有分化能力 .将紫红色愈伤组织转入 MS补加 0 .5~ 4 .0 mg/ L 6-BA、0 .2~1 .0 mg/ L NAA的培养基上 ,经低温 -4℃和 4℃处理 1 0 d,然后置于 2 5℃下光照培养 ,2周后就能诱导出密集的丛生芽 .不定芽转接到 MS+0 .5~ 1 .0 mg/ L IAA+0 .1 mg/ L 6-BA的培基上 ,经 -4℃低温处理 2周 ,然后在 2 5℃下培养 3周就可生根 .低温处理是水母雪莲愈伤组织再分化的关键条件 .组织细胞学观察表明 ,水母雪莲的器官发生主要为不定芽途径 .

【Abstract】 Leaf segments of Saussurea medusea Maxim. were cultured on MS medium supplemented with 0.2~4.0 mg/L NAA, 0.2~4.0 mg/L 6 BA. Three weeks later callus foration would be induced from the excised leaves. An induced frequency of 100% was obtained on the MS medium with 2.0 mg/L NAA +.02 mg/L 6 BA. With subculture, three types of callus were separated from primary callus. They presented an obvious difference in re differentiation ability. The white and the light greenish yellow callus didnt differentiate. Only the purplish red callus had differentiation capability. The purplish red callus were shifted to MS medium supplemented with 0.5~4.0 mg/L 6 BA, 0.2~1.0 mg/L NAA. After they were treated under low temperature (-4℃ and 4℃)for ten days, they were cultured at 25℃ in light for two weeks. As a result, a large number of adventitious buds were regenerated. The adventitious buds were cultured on MS medium with 0.5~1.0 mg/L IAA +0.1 mg/L 6 BA under low temperature (-4℃) for two weeks. Then, roots would occur after they were cultured at 25℃ for three weeks. The low temperature was a key factor on re differentiation of callus of S.medusa Maxim.. The action of low temperature in organogenesis of S.medusa Maxim. would be probably similar to the vernalization in seeds. Using cytohistological method, we observed that the regeneration of adventitious buds was the main way of organogenesis of S.medusa Maxim.
