高效生物杀虫剂BtA化学耦合技术研究
Study on the technique for conjugating Bacillus thuringiensis \|crystal toxin with Abamectin
【摘要】 提出了苏云金芽胞杆菌毒蛋白与阿维菌素化学耦合的方法 ,实现了多位点生物杀虫毒素 Bt A的工业化生产。认为 Bt A的化学耦合 ,一方面是将 Bt的伴胞晶体进行酶切改造 ,形成带末端氨基的原毒素 ,另一方面是将放线菌的杀虫毒素阿维菌素通过对 C2 3 位置上的羟基进行激活、衍生化 ,形成带羧基的阿维菌素衍生物 ,最后利用氨基 -羧基耦联剂 EDC进行耦合 ,实现单体双生物毒素 Bt A的结构改造。高效生物杀虫剂 Bt A的研究成功 ,使其杀虫谱扩大为五个目 (鳞翅目、同翅目、双翅目、鞘翅目和螨类 ) ,杀虫速率提高为 >85 %死亡率 /2 4小时 ,保持了无公害的特性 ,对于农业环保具有重要意义
【Abstract】 The present paper dealt with the technique for conjugating Bacillus thuringiensis Crystal toxin with Abamectin. In the process of chemical conjugation,the Bt\|crystal toxin was dissolved by DTT(dithiothreitol) to form the toxin protein. Abamectin was changed by a series of chemical reaction of carboxylation to form carboxy\|abamectin. The toxin protein was conjugated with the carboxy\|abamectin under the treatment of EDC \[1\|Ethyl\|3\|(3\|dimethylaminopropyl) carbodiimide Hydrochloride)\]to form BtA biocide. The bioassay of BtA was conducted to display that BtA biocide had a wide range and high killing speed against pests including Lepidoptera,Homoptera,Diptera,Coleoptera and Mite from agricultural crops. The successful formation of BtA biocide has a great significance in biological control and agricultural environment protection.
