【摘要】 目的 :研究D15S657的多态性与胰岛素依赖型糖尿病(IDDM)和成人迟发型IDDM(LADA)的关系。方法 :应用PCR -PAGE结合产物直接测序的技术 ,在105名无亲缘关系的成都地区正常汉族人 ,48名IDDM患者以及22名LADA患者中 ,对IDDM易感位点IDDM3定位区域内的遗传标记D15S657的多态性进行分析。结果 :D15S657位点的等位基因A5的频率在IDDM组和LADA组中显著高于正常对照组 ,分别为0 1875∶0 0619 ,P=0 0011 ,Pc=0 008和0 2045∶0 0619 ,P=0 0055 ,Pc=0 0385。结论 :D15S657与中国人群的IDDM和LADA相关 ,IDDM与LADA发病具有相同的遗传因素

【Abstract】 Objective:TostudythepolymorphismofgeneticmarkerD15S657locusanditsassociationwith insulin -dependentdiabetesmellitus(IDDM)andlatentauto -immunediabetesinadults(DADA).Methods:Poly merasechainreactionpolyacrylamidegelelectrophoresis(PCR -PAGE)anddirectsequencingofPCRproductswere usedtoanalyzethepolymorphismofD15S657olcus,ageneticmarkerintheolcalizationregionofinsulin -depen dentdiabetesmellitus3(IDDM3),in105normalpeopleofChineseHannationalityand48PatientswithLADA. Results:ThefrequencyofalleleA5atD15S657locusinpatientswithIDDMandpatientswithLADAwassignifi cantlyhigherthanthatinthecontrolgroup.Theirfrequencieswere0.1875∶0.0619(P=0.0011,Pc=0.008)in IDDMgroupand0.2045∶0.0619(P=0.0055,Pc=0.0385)inLADAgroup.Conclusions:D15S657isnotonly associatedwithIDDM,butalsowithLADAinChineseHannationalitypopulation.IDDMandLADAhavethesame geneticfactors.Polymorphismo
