中药红曲基原真菌的高效毛细管电泳色谱法鉴别

【摘要】 目的:建立一种中药红曲基原真菌新的鉴定方法。方法:采用毛细管区带电泳分离模式,背景电解质为20 m m ol/L硼酸盐缓冲液(pH 9.11),电压20kV,温度25 ℃;紫外检测波长214 nm ,压差进样,进样时间5 s。结果:不同种红曲霉胞内和胞外水溶性蛋白质的成分和含量均存在显著差异。根据红曲霉胞内或胞外水溶性蛋白质4~10 m in 区间内的指纹图谱及这些峰的相对含量,可以有效地鉴别红曲霉属真菌,结果与形态学以及扫描电镜的检验结果一致。结论:高效毛细管电泳可用于红曲霉等真菌的分类和鉴定。

【Abstract】 Objective: To establish a new method to identify original fungi of Chinese traditional medicine: Hongqu . Methods: The capillary zone electrophoresis method was used. The buffer used for protein separation was 20 mmol/L borate of pH 9.11. Proteins were monitored at 214 nm, voltage at 20 kV and pressure injection 5 s. The temperature of the capillary during electrophoresis was kept constant at 25℃. Results: The number of peaks and the contents either extracellular proteins of Monascus or intracellular proteins was obviously different, we could distinguish easily Monascus by the fingerprint peaks within 4 and 10 minutes either of extracellular proteins or of intracellular proteins. The results were consistent with morphologic study and analysis with scanning electron microscope. Conclusion: HPCE assay technique can classify and identify of Monascus fungi.
