甲硫氨酸脑啡肽对NIH3T3细胞和人肝癌Bel 7402细胞生长具有抑制作用

【摘要】 目的 :研究甲硫氨酸脑啡肽对体外培养的NIH3T3细胞、人肝癌Bel740 2细胞增殖的影响。方法 :MTT微培养染色法检测不同质量浓度的甲硫氨酸脑啡肽对NIH3T3细胞、人肝癌Bel740 2细胞的作用以及一定质量浓度的纳洛酮、阿片δ受体单克隆抗体、5 氟脲嘧啶对这种作用的影响。结果 :(1) 10mg·L-1至 80mg·L-1的甲硫氨酸脑啡肽对这 2种细胞均有不同的抑制效应 (NIH3T332 %~ 6 6 %与Bel740 2 36 %~ 6 4% ) ;(2 )纳洛酮能阻断甲硫氨酸脑啡肽对细胞抑制效应 ,阿片δ受体单克隆抗体没有这种阻断能力 ;(3) 5 氟脲嘧啶 (10mg·L-1)和甲硫氨酸脑啡肽 (5 0mg·L-1)有协同作用。结论 :甲硫氨酸脑啡肽能抑制细胞增殖。提示NIH3T3细胞和Bel 740 2细胞胞膜上可能存在阿片 ζ受体介导。

【Abstract】 Objective: To investigate the effect of methionine enkephalin on the proliferation of cultured NIH3T3 cells and human hepatoma Bel7402 cells. Methods: Utilizing the MTT microculture tetrazolium dye, the effect of synthesized methionine enkephalin at various concentrations on the growth of NIH3T3 cells and human hepatoma Bel 7402 cells was assessed. The effects of naloxone, monoclonal antibody to opioid δ receptor, 5 Fu on the action of methionine enkephalin were also observed. Results: (1) Methionine enkepaalin (10 mg·L -1 to 80 mg·L -1 ) significantly inhibited the proliferation of the two kinds of cells (NIH3T3 32% 66%, Bel 7402 36% 64%). (2) Naloxone could block the activity of methionine enkephalin, but the monoclonal antibody to opioid δ receptor had no such ability.(3) 5 Fu (10 mg·L -1 ) had synergistic inhibitory effect with methionine enkephalin (50 mg·L -1 ). Conclusion: Methionine enkephalin can inhibit the proliferation of the two kinds of cells. It is possible that opioid ζ receptor exists on the cellular membrane of NIH3T3 cells and Bel7402 cells.
