氨甲喋呤标记抗体导向治疗自身免疫性血小板减少性紫癜的体外实验研究
Study on targeting therapy of idiopathic thrombocytopenic purpura with MTX-labelled antibody in vitro
【摘要】 目的 :将静脉注射用免疫球蛋白 (IVIG)与免疫抑制剂交联制备成标记抗体 ,使之对单核 -巨噬细胞产生特异的杀伤作用 ,为其临床治疗自身免疫性血小板减少性紫癜 (ITP)奠定实验基础。方法 :用间接交联法将 IVIG与 MTX制备成标记抗体 ,用间接免疫荧光法检测标记抗体的Fc段结合活性 ,以 Fc受体阳性的小鼠巨噬细胞和人单核细胞白血病细胞株 U937为靶细胞 ,用MTT法测定标记抗体的杀伤活性。结果 :标记抗体对巨噬细胞的杀伤作用明显大于游离 MTX;标记抗体对 Fc受体阳性细胞的杀伤作用明显大于 Fc受体阴性细胞。结论 :标记抗体在体外对单核 -巨噬细胞显示了高度特异的杀伤活性
【Abstract】 ? Objective:To conjugate IVIG and MTX to produce a specific cytotoxicity upon phagocytes.Methods:MTX was conjugated with IVIG by indirect conjugating methods.HSA was used as an intermediary to conjugate MTX with IVIG.The indirect immunofluorescence was adopted to test the binding ability of Fc fragment.MTT assay was used to measure the cytotoxicity of conjugation on phagocytes.Results:Conjugation showed stronger cytotoxicity upon target cells than free MTX,and it showed only less cytotoxic effect on Fc receptor negative cells compared with the positive ones.The specific cytotoxicity of IVIGHSAMTX was significantly stronger than that of MTX.Conclusion:In vitro the conjugation showed a highly specific cytotoxicity upon phagocytes.
