镉对免疫系统毒性的肾上腺素能受体信号传导机理

【摘要】 应用普通动物建立心得安、酚妥拉明及两者联合作用的动物模型,观察不同剂量镉对大鼠免疫系统的影响及与肾上腺素能受体信号传导的关系。结果表明:镉主要蓄积于肝、肾、肾上腺,并引起血浆肾上腺素水平改变,使淋巴细胞cAMP 降低以及T 淋巴细胞增殖功能和巨噬细胞吞噬功能受抑制;用心得安阻断β- 受体后,仅巨噬细胞吞噬率受影响;用酚妥拉明阻断α- 受体后,NE、E、T 细胞和巨噬细胞均受影响;α- 受体、β- 受体同时阻断后,仅中、高镉组CD+4 /CD+8 降低。提示镉免疫毒性的β- 受体- cAMP机理确实存在

【Abstract】 The animal models of Propranol and Phentolamine were used to observe the effects of different dozes of cadmium (CdCl 2)on immune system and the relationship with adrenoceptor signal transduction.The results showed that ,cadmium accumulated in liver,spleen,adrenal gland and caused change of adrenalin in serum toghether with decrease of cAMP,inhibition of T lymphocyte proliferation and macrophage phagocytic function.There was no significant difference in all indexes except phagocytic rate of macrophage when β adrenoceptor waw blocked by propranol.On the other hand,when the α-adrenoceptor was blocked by phentolamine,significant differences existed in noradrenalin,adrenalin,T lymphocyte proliferation and macrophage phagocytic function.In the animal models with blocked α-,β-adrenoceptors,only CD + 4/CD + 8 in group of mid and high dozes of cadmium was decreased.Our research indicated that adrenoceptor signal trancduction existed and had a very important role in mechanism of cadmium’s immunotoxicity.
