多聚酶链反应检测结核杆菌复合群方法的建立及临床应用

【摘要】 采用多聚酶链反应 （ＰＣＲ） 对结核杆菌复合群基因组进行扩增， 得到一２８５ ｂｐ ＤＮＡ条带。经Ｓｏｕｔｈｅｒｎ 印迹证实为特异性扩增。用ＰＣＲ方法对临床４０ 份待测标本进行检测，并与结核杆菌常规培养以及高渗培养等方法进行比较， 其阳性率分别为６０．０％ 、７．５％ 和０％ ， 差异均有显著性意义 （均为Ｐ＜ ０．０１）。

【Abstract】 A polym erase chain reaction (PCR) assay for the rapid detection of M.tuberculosis com plex w as developed. The PCRsystem w as used in the identification ofm em bers ofthe M.tuberculosis com plex and the specific productofam plification is a 285 bp DNA fragm entconfirm ed w ith southern blot. The 40 clinical specim ens w ere detected w ith the m ethods of PCR, M.tuberculosis culture, L-form of M.tuberculosis culture and acid-fast staining, at the sam e tim e, w ith the positive rate being 60.0 % , 7.5 % , 0 % , respectively.
