HIV-1型衣壳蛋白P24在大肠杆菌中的表达、纯化及鉴定

【摘要】 用聚合酶链式反应 (PCR)从 HIV - 1gag基因序列中扩增出衣壳蛋白 (P2 4)基因 ,插入表达载体p GEX- 4T3中 ,构成重组质粒 p GEX- p2 4,在大肠杆菌 BL 2 1中高效表达出重组 P2 4。经 Glutathione- Sepharose4B亲和层析纯化后 ,重组 P2 4在间接 EL ISA和免疫印迹中表现出很高的抗原特异性和免疫反应性。

【Abstract】 A recombinant plasmid(pGEX p24) was constructed by inserting the capsid protein gene P24 amplified from HIV 1 gag genome by PCR into a expression vector pGEX p24 and transfered to E.coli. After being purified by Glutathione Sepharose 4B affinity chromatography, the expressed P24 was detected by ELISA and Western blot and showed good antigenic specificity and immunoreactivity.
