戈米辛J对低密度脂蛋白氧化修饰的抑制作用
Inhibitory Effect of Gomisin J on the Oxidative Modification of Low Density Lipoprotein
【摘要】 目的研究戈米辛J对铜离子和内皮细胞诱导的低密度脂蛋白氧化修饰的作用。方法以巴比妥酸反应物质、荧光物质及电泳迁移率测定低密度脂蛋白的氧化程度。结果戈米辛J抑制铜离子诱导的LDL氧化修饰,有效质量浓度为5μmol/L,IC50(95%可信限)为26.2(16.5~41.5)μmol/L,尚可抑制氧化的低密度脂蛋白产生的荧光物质及琼脂糖凝胶电泳迁移率;对内皮细胞诱导的低密度脂蛋白氧化修饰,戈米辛J的抑制作用呈剂量依赖关系。结论戈米辛J对铜离子和内皮细胞诱导的低密度脂蛋白氧化修饰有显著抑制作用。
【Abstract】 Purpose To study the effect of gomisin J on the oxidative modification of low density lipoprotein(LDL) induced by copper and endothelial cells(EC). Methods Oxidative modification of LDL was examined by thiobarbituric acid reactive substances(TBARS),lipofusion and electrophoretic mobility. Results Gomisin J inhibited copper induced LDL peroxidation with IC 50 (95% confidence limits)26.2(16.541.5) μmol/L。It could also inhibit the products of lipofusion and the rate of eletrophoretic mobility of oxidized LDL (ox LDL).Additionally,gomisin J showed an inhibitory effect on EC induced ox LDL in a dose dependent manner. Conclusions Gomisin J inhibits the oxidative modification of LDL induced by copper and EC.
