【摘要】 为探讨髓细胞性白血病（ｍ ｙｅｌｏｉｄ ｌｅｕｋｅｍ ｉａ）患者Ｐ－１７０ 蛋白过度表达与预后的关系，用免疫组化ＬＳＡＢ法（ｌａｂｅｌｌｅｄ ｓｔｒｅｐｔａｖｉｄｉｎ ｂｉｏｔｉｎ ａｓｓａｙ）检测６６ 例急性髓细胞性白血病（ａｃｕｔｅ ｍ ｙｅｌｏｉｄ ｌｅｕｋｅｍ ｉａ，ＡＭＬ）及２０ 例慢性髓细胞性白血病（ｃｈｒｏｎｉｃ ｍ ｙｅｌｏｉｄ ｌｅｕｋｅｍ ｉａ， ＣＭＬ）患者外周血单个核细胞的Ｐ－１７０ 蛋白表达。结果表明，初治组ＡＭＬ患者Ｐ－１７０ 表达率为２６．５％ （９／３４）；难治／复发组为６８．４％ （１３／１９），比初治组明显增高（Ｐ＜ ０．００５）；完全缓解组为２３．１％ （３／１３），与初治组差异无显著性。诱导化疗Ｐ－１７０＋ 者完全缓解（ＣＲ）率为２２．２％ （７／９），Ｐ－１７０－ 者为８０％ （２０／２５），二者差异有显著性（Ｐ＝ ０．００３６）。ＣＭＬ慢性或加速期８ 例患者无１ 例Ｐ－１７０ 表达，急变期患者Ｐ－１７０ 表达率为５０％ （６／１２），二者差异有显著性（Ｐ＝ ０．０４２）。提示Ｐ－１７０ 蛋白过度表达是髓细胞性白血病化疗耐药和不良预后的标志更多还原

【Abstract】 Labelled streptavidin biotin immunohistochemical staining technique(LSAB) was used to detect the mdrl associated P glycoprotein (P gp) expression in 66 patients with AML and 20 with CML. In AML patients, 9/34 at diagnosis, 13/19 in relapses or refractory diseases, and 3/13 in CR showed P gp overexpression. The CR rates of induction therapy were 20/25(80%) in P gp - and 2/9(22.2%) in P gp + cases(P= 0.0036 ). P gp was also detected in 8/16 CML cases in blast crisis, while none in eight chronic or accelarative cases, P=0.042. P gp overexpression may contribute to drug resistance and poor prognosis in myeloid leukemias.更多还原
