神经生长因子及其受体在过量维生素A酸所致神经管畸形发生中的作用

【摘要】 用免疫组织化学方法研究了神经生长因子(NGF)及其受体(trkA)在正常金黄地鼠和过量维生素A 酸致畸金黄地鼠胚胎神经管及其周围间充质的分布状况。结果显示:NGF广泛分布于正常神经上皮及其内、外界膜和周围间充质中;实验组在服药后46h,上述各部位NGF和trkA 含量均明显减少。表明NGF和trkA 与神经管正常发生密切相关,其含量减少可能是过量维生素A酸致神经管畸形的机制之一

【Abstract】 Both malformed embryos induced by excess retinoic acid and normal ones of golden hamsters were studied on the expression of nerve growth factor(NGF) and its receptor(trkA)in the neural tubes and their surrounding mesenchyme. The results were as follows: the NGF and trkA were extensively expressed in the neuroepithelia, internal and external limiting membranes and their surrounding mesenchyme in normal groups. Otherwise, the content of both NGF and trkA in the corresponding regions of the experimental groups were obviously reduced, especially 46 hours after administration. The results suggest that NGF and trkA have a close relation with the development of the neural tube and quantitative decrease of both NGF and trkA may be one of the mechanisms by which excessive retinoic acid induces neural tube defects.
