【摘要】 目的　探讨在胃癌组织中 Ｐ Ｔ Ｅ Ｎ 基因和蛋白的表达情况及临床病理意义。方法　应用免疫组织化学和原位杂交技术检测５０ 例胃癌及其癌旁组织中 Ｐ Ｔ Ｅ Ｎ 蛋白及 Ｐ Ｔ Ｅ Ｎ ｍ Ｒ Ｎ Ａ 表达情况，同时结合病人的临床病理资料进行分析。结果　５０ 胃癌组织中， Ｐ Ｔ Ｅ Ｎ 蛋白阳性表达率４８ ％ ，阳性率５２ ％ ，阳性反应分布在胞浆中。５０ 例胃癌组织中 Ｐ Ｔ Ｅ Ｎ ｍ Ｒ Ｎ Ａ 阳性表达率６０ ％ ，阳性率４０ ％ ，阳性信号显示于胞浆中。 Ｐ Ｔ Ｅ Ｎ 蛋白及 Ｐ Ｔ Ｅ Ｎ ｍ Ｒ Ｎ Ａ 在胃癌组织中的阳性表达与患者性别，年龄无明显相关性，但均与组织分化程度明显相关，高分化腺癌组的阳性率分别为７７ ．８ ％ 及８８ ．９ ％ ，低分化腺癌组的阳性率分别为３１ ．３ ％ 及４３ ．８ ％ 。结论　胃癌组织中存在较高比例的 Ｐ Ｔ Ｅ Ｎｍ Ｒ Ｎ Ａ 及 Ｐ Ｔ Ｅ Ｎ 蛋白阳性表达，说明在胃癌的发生发展中， Ｐ Ｔ Ｅ Ｎ 基因失活起着重要作用，两者的阳性表达均有一定的预后意义。更多还原

【Abstract】 Objectives To investigate the expression of PTEN gene in cacinoma of stomach and its clinicopathological.Methods Paraffin embedded tissues from 50 patients with cacinoma of stomach were analysed for PTEN gene expression by immunohistochemical methods and in situ hybridization technique. Results Of the 50 carcinoma of stomach cases, 24 (48%) were positive for PTEN protein, 26(52%) lacked PTEN protein, the positive signals were seen in the cytoplasm; 30(60%) were PTEN mRNA positive, 20(40%) lost PTEN mRNA and PTEN mRNA was clearly observed in the cytoplasm of carcinoma ttissues.PTEN expression did not show correlation with such factors as sex, age in caxcinoma of stomach, but there was a significant difference between the pattern of expression of PTEN in well differentiated adenocarcinoma and poorly differentiated adenocarcinoma (P<0.05). Conclusions There was a higher incidence of negative expression of PTEN mRNA and protein in carcinoma of stomach, which indicates that PTEN gene inactivation may play an important role in the genesis and development of some carcinoma of stomach .The positive may be features for favorable prognosis in patients with carcinoma of stomach更多还原
