自制损伤机建立猫弥漫性轴索损伤模型
Preparation of cat diffuse axonal injury model with self-made injury device
【摘要】 目的:观察猫头颅瞬间侧向旋转引起脑弥漫性轴索损伤(DAI)模型的稳定性,并探讨损伤的机制。方法:对照组4只、损伤组38只,采用自制弥漫性轴索损伤装置,将损伤组猫头颅于11~12ms内在冠状面测向旋转60°造成剪力伤,于伤后6、12、24、48、72、120、144、168h分批处死动物,制脑切片、行嗜银及HE染色、光镜下观察神经轴索变化.结果:伤后即刻猫意识均丧失.6只30min内死亡,肉眼亦见蛛网膜下广泛出血,其余存活猫在伤后6~168h,肉眼见蛛网膜下腔广泛出血,光镜下见脑干、胼胝体、大脑脚等部位的神经轴索有不同程度的肿胀、断裂、轴索回缩球形成、成簇小胶原细胞增生等征象.结论:本研究成功地建立了猫头颅侧向瞬间旋转脑弥漫性轴索损伤模型,表明剪力可引起轴索损伤,为DAI进一步研究提供了条件.
【Abstract】 AIM: To observe and explore the stability of the self made model and the mechanism of neuronal axons injury in cat brain by rolating its head. METHODS: 38 cats were rolated quickly using self-made injury model for 11~12ms in the coronal plane by 60°. Then those injured cats were killed at 6, 12, 24, 48. 72, 120, 144 and 168 hours respectively. Tisssue slides of the brain were made and processed with silver and with HE slaining and observed under microscope. RESULTS:Compared with control group (n=4), all injuried rats lost consciousness after injury.among which six cats died of diffose subarachnoid hemmorage in 30 minute.The survival cats were seen grossly with diffose subarachnoid hemmorage and seen under microscope with the signs of various severity of axonal swelling, discontinuity and axonal retraction ball at the brainstem, corpus collosum and cerbral peduncle.CONCLUSIONS: The present study successfully prepareds a cat model of brain DAJ by quickly rolating injury sugge sting that the shearing force may lead to DAI and the further DAI studies should be needed.
