白细胞介素-1受体拮抗剂对体外培养的大鼠大脑皮层神经细胞缺氧性损伤的影响及其基因转染治疗

【摘要】 目的:观察IL1的受体拮抗剂(IL1ra)对缺氧性神经细胞的影响,探讨IL1在神经细胞缺氧损伤中的作用以及基因转染治疗的可能性。方法:(1)以缺陷型HSV为载体构建的pHSVIL1ra转染体外培养的神经细胞为实验组,以pHSVLacZ转染的体外培养的神经细胞为对照,进行体外缺氧实验。以神经细胞的3HGABA和3HGlu摄取能力及细胞凋亡情况为判定损伤程度的指标。结果:转染pHSVIL1ra的神经细胞缺氧后,其3HGABA和3HGlu的摄取较转染pHSVLacZ的神经细胞有所增高,神经细胞凋亡的程度少于对照。结论:pHSVIL1ra转染可减轻缺氧引起的神经细胞急性损伤和迟发性损伤。

【Abstract】 Objective: To observe the effect of IL 1 receptor antagonist (IL 1ra) on the hypoxic damage of cultured neurons and study the possible treatment with experimental gene transfection. Methods: Hypoxic experiment was done in the experiment group (transfected neurons with pHSV IL 1ra) and the control group (transfected neurons with pHSV LacZ). The markers of hypoxic damage included the uptake ability of the neurons for 3H GABA and 3H Glu and the cell apoptosis. Results: After hypoxia, uptake ability of the 3H GABA and 3H Glu were higher in experiment group than in the control. Apoptotic cells were fewer in the experiment group than in the control. Conclusion: Transfection of pHSV IL 1ra can reduce both the acute and the delayed hypoxic neurons damage.
