【摘要】 目的：主要探索Ｔ－２毒素对体外培养软骨细胞致损临界值以及硒保护作用。方法：应用单层培养传代软骨细胞，用生化方法检测软骨细胞ＤＮＡ量和蛋白量，透射电子显微镜观察软骨细胞超微结构。结果：（１）Ｔ－２毒素在０．００１～０．００５ｍｇ／Ｌ培养液中（以下相同）剂量范围内，随浓度增加，Ｔ－２毒素对软骨细胞ＤＮＡ合成和细胞分裂增殖影响逐渐增大，加硒后该影响有所减弱，但仍不能阻止损伤的发生；（２）Ｔ－２毒素在０．０００５ｍｇ／Ｌ时，对软骨细胞的影响甚微；Ｔ－２毒素在０．００１ｍｇ／Ｌ和０．００５ｍｇ／Ｌ时，细胞损伤明显，与对照组比较有显著差别（Ｐ＜０．００１）。更多还原

【Abstract】 In the present investigation,we observed the critical value of the damage of T2 toxin to chondrocytes and the protection of Se.The amounts of DNA and protein in the singlelayer cultured chondrocytes were assessed by the biochemical methods and the transmission electron microscope was utilized to observe the ultrastructure of chondrocytes.The results showed:(1) T2 toxin could restrain both the DNA synthesis and the cell proliferation of chondrocytes in the media containing different concentrations of T2 toxin (0.001￣0.005mg/L).The effect of T2 toxin on the chondrocyte damage was increased with the concentration of T2 toxin in the given does.When Se was added into the media,the chondrocyte damage by T2 toxin was decreased,but can not prevented.(2)T2 toxin led to few effects on chondrocytes as its concentration was 0.0005mg/L.It caused evident damage to chondrocytes when its concentration was 0.001mg/L and 0.005mg/L and the cell damage was significantly different from that in the control group (P<0.001).更多还原
