【摘要】 HPV16与宫颈癌的发生发展关系密切。其基因功能区包括早期区（E区）、晚期区（L区）及调节区。E区与细胞转化、致癌有关；L区分为L1和L2，L1区主要编码病毒衣壳蛋白，表达的蛋白能刺激机体产生保护性抗体，因此通过对HPV16L1基因的克隆及表达，可为制备基因工程疫苗和诊断试剂盒打基础。本实验通过甲醇诱导培养法及SDS－PAGE电泳对本室已立了HPV16L1－ρPIC3．5／GS115重组菌株进行了筛选，获得了1株HPV16L1晚期基因的GS115酵母菌重组表达株，所表达的蛋白质分子量为80KD。与预计相同，并证明第3天时L1蛋白的表达量最大。更多还原

【Abstract】 HPV 15 is in clos relation to the occuring and developing of cervical cancer. Its gene function restions include : E region and L region. L region includes L1 and L2. L1 region codes virus capsid protein. This protein can induce the body to generate protective antibodies. So, We can make the basis of producting gene vaccine and diagnosis kit by clonint and expressing HPV 16 L1 gene.In this project, the HPV 16 L 1 -pPIC3. 5/GS115 recombinant trains which can express L1 protein have been screened by methanol induction and SDS -PAGE. It has been tested that the protein’ s molecular weight is 80KD, just as expected. and that the amount of expression on the 3nd day is the largest of six days.更多还原
