癌胚抗原真核表达质粒构建与鉴定
Construction and Identification Of Eukaryotic Expression Plasmid Encoding Human Carcinoembryonic Antigen
【摘要】 癌胚抗原(CEA)是一种重要的肿瘤相关抗原,同时也是针对某些肿瘤治疗的有效靶抗原。为了探讨运用表达CEA的重组质粒进行抗肿瘤治疗的可行性,本研究构建了两套表达CEA的真核重组质粒。方法:运用基因工程技术将编码人CEA的全长2.4kbcDNA分别插入到真核表达载体pcDNA3及VR1012中,并用多种限制性内切酶对重组质位进行酶切鉴定。结果:构建的重组质粒均含有24kbEA片段,且插入pcD-NA3及VR1012中的2.4kb片段的方向均为正向,将这两套重组质粒分别命名为pCEA-1及pCEA-2。结论:本研究结果为进一步研究表达CEA的重组质位的免疫效应打下了基础。
【Abstract】 Carcinoembryonic antigen (CEA)is an important tumor associated antigen and an effective target antigen for the therapy of several malignant malignant . In order to investigate the anti -- tumor effects of recombinant plasmids s expressing CEA, eukaryotic expression plasmids containing CEA cDNA were constructed. Methed: Genetic engineering techniques were applied to inserting a 2.4gb fragment encoding full -- length human CEA into two eukaryotic expression vectors pcDNA3, VR1012 respectively . The resultant recombinant plasmids were cleaved with appropriate endonucleases. Results: The insertion 2. 4kb DNA fragment was certified to be in the right orientation and the two recombinant plasmids were named pCEA -- 1, pCEA -- 2 respectively. Conclusions: The results paved the way for future research on the immunological effects of the two recombinant plasmids expressing CEA.
