【摘要】 由于常规福尔马林固定和石蜡包理过程造成的抗原封闭与结构改变，限制了一些抗体在免疫组织化学中的应用。为了恢复组织的抗原性，提高免疫组织化学的检测阳性率，我们应用高压锅，微波炉和胰蛋白酶分别对石蜡切片进行处理。结果显示，一些抗体过去只能用于冰冻切片，经前处理后可用于石蜡切片，一些抗体的敏感性增强，经高压锅处理的免疫染色重复性较微波炉处理得好，个别抗体适用于酶消化。总之，在免疫组织化学染色过程中应根据抗原抗体的特性选用适当的前处理方法。更多还原

【Abstract】 The application of some antibodies always has been problematic due to routine formalin fixation and paraffin embedment. In order to unmask the antigens of tissue sections and increase the incidence of immunohistochemical positive results, pressure cooker, microwave and trypsin were used respectively to pretreat paraffin embedded tissue sections. The results showed that (1) some antibodies used to be applied on frozen sections can be used on paraffin sections after processing of pressure cooker and microwave, (2) the sensitivity of some antibodies has been enhanced, (3) the repetition of staining in sections pre-treated by pressure cooker was better than that pretreated by microwave and a few antibodies were suitable for enzyme digestion. In brief, according to the property of antigens and antibodies, a proper pre-treatment processing method should be selected in the course of immunohistochemistry.更多还原
