表皮生长因子与谷氨酰胺对大鼠乙酸性结肠炎的影响
Effect of epidermal growth factor and glutamine on acetic acid -induced colitis in rats
【摘要】 本研究通过对大鼠结肠粘膜组织学观察,过氧化脂质(LPO)、髓过氧化物酶(MPO),谷胱甘肽过氧化物酶(GSH—Px)及其线粒体ATPase水平的检测,旨在探讨表皮生长因子(EGF)和谷氨酰胺(GLN)防治大鼠乙酸性结肠炎的机理.结果表明:(l)EGF组、GLN组及EGF+GLN组结肠粘膜损伤指数均明显低于乙酸组(P<0.01);光镜、透射及扫描电镜观,EGF+GLN组结肠粘膜炎症明显轻于其余3组。(2)EGF组、GLN组和EGF+GLN组LPO、MPO水平分别明显低于乙酸组;4组间GSH—Px活性差别无显著性(P>0.05);GLN组Na+—K+—ATPase、Ca2+—ATPase活性均显著高于乙酸组,其Ca2+—ATPase活性还明显高于EGF组;EGF+GLN组Na+—K+—ATPase、Ca2+—ATPase和Mg2+—ATPase水平均显著高于乙酸组,该组后两种ATPase还明显高于EGF组。提示EGF与GLN联合应用,可增加组织营养、能量供给,抑制脂质过氧化损伤,防治炎症,从而对结肠粘膜具有明显的保护作用。
【Abstract】 In order to study the cytoprotection of epidermal growth factor (EGF) and glutamine(GLN) on acetic acid -induced colitis in rats, colonic pathomorphology and levels of lipid peroxides (LPO), myeloperoxidase(MPO), glutathione peroxidase(GSH -Px)and ATPase of mitochondria were observed. The results showed that:l. The damaged index of colonic mucosa was significantly lower in EGF group, GLN group, EGF + GLN group than that in acetic acid group(P<0.01), the colonic inflammation was obviously less severe in EGF +GLN group than in other three groups observed under macroscope, light microscope, transmission electron microscope and scanning electron microscope; 2. The concentration of LPO and MPO were very much lower in EGF group, GLN group, EGF + GLN group than in acetic acid group. While there was not much difference of GSH-Px between these four groups (P>0.05). The contents of Na+-K+-ATPase, Ca2+ -ATPase were significantly higher in GLN group than in acetic acid group, and Ca2+- ATPase of GLN group was also higher than EGF group. The activities of Na + -K+ -ATPase, Ca2+-ATPase, Mg2 + -ATPase were obviously higher in EGF + GLN group than in acetic acid group, Ca2+ -ATPase, Na+ -K+ -ATPase levels of EGF + GLN group were also higher than EGF group. It is suggested that EGF combined with GLN has strong protective effect on colonic mucosa by increasing supplies of nutrition, energy and inhibited damages of lipid peroxidation which prevent inflammation and accelerate the healing of tissue lesion.
