日本血吸虫中国大陆株23kDa膜蛋白的基因克隆与序列分析
GENE CLONING AND SEQUENCING OF THE VACCINE CANDIDATE MEMBRANE PROTEIN Sj23 OF SCHISTOSOMA JAPONICUM CHINESE STRAIN
【摘要】 本研究根据曼氏血吸虫疫苗候选分子Sm23的基因序列,设计了一对引物,并通过聚合酶链反应(PCR)基因克隆技术,从日本血吸虫中国大陆株cDNA文库中克隆出了中国大陆株23kDa膜蛋白基因。DNA序列分析表明,编码日本血吸虫中国大陆株Sj23膜蛋白的基因含有657bp,与曼氏血吸虫Sm23的核着酸有79.5%的同源性,而与日本血吸虫菲律宾株的Sj23的同源性为100%。该研究为发展Sj23多肽疫苗奠定了基础。
【Abstract】 A pair of primers was synthesized according to the sequence of Sm23,and the gene of the 23 kDa membrane protein of S. japonicum Chinese strain was cloned from the cDNA library of this strain through polymerase chain reaction (PCR) technique with the primers. The results of DNA sequencing of the gene showed that encoding Sj23 sequence included 657 bp,which was 79. 5 % in homology with Sm23,moreover,it was 100% in homology with Sj23 of S. joponicum philippine strain.
