Co-TPPS4清除氧自由基和过氧化氢的双功能作用
Dual Functions of Co-TPPS4 in Scavenging Oxygen Free Radicals and Hydrogen Peroxide
【摘要】 以四-(对-磺酸基苯基)钴卟啉[Co-TPPS4]作为超氧化物歧化酶(SOD)和过氧化氢酶(CAT)双功能模拟酶,研究其仿SOD和CAT活性及抗脂质过氧化活性。结果应用核黄素-蛋氨酸光照法证实其在10-5~10-6mol/L浓度范围内有清除超氧阴离子自由基的作用;分光光度法证实其能催化过氧化氢的分解,当其浓度由5μmol/L增至30μmol/L时,分解率由11.9%增至62.4%;小鼠肝匀浆法测定Co-TPPS4、SOD、CAT,均表明具有较好的抗脂质过氧化的作用。
【Abstract】 In this article,the tetraphenylporphinesulfonatocobalt (Co-TPPS4) was used as dual function mimicry of superoxide dismutase (SOD) and catalase (CAT). The first function, scavenging O2, has been proved by using riboflavin-methionine photoreduction method in the concentration range of 10-5~10-6 mol/ L. The second function, scavenging H2O2, has been demonstrated by means of spectrophotometry. When the concentration of Co-TPPS4 increased from 5 μmol/L to 30 μmol/L, the decomposing rate of H2O2 increased from 11.9% to 62. 4%. Co-TPPS4, SOD and CAT were measured by the liver homogenate technique of mice and the results showed that they had obvious action of decreasing the lipid peroxidation.
