三对水稻抗白叶枯病近等基因系的RAPD分析
RAPD Analyses of Three Nearisogenic Lines of Rice Resistant to Bacterial Blight
【摘要】 以水稻抗白叶枯病3对近等基因系(IR24与IRBB3、IR24与IRBB4和IR24与IRBB5)为材料,对近等基因系基因组DNA进行RAPD分析,结果显示:经120个10-核苷酸随机引物对3对近等基因系基因组DNA的PCR扩增,有2个引物(OPK-11和OPN-04)在IRBB3中各扩增出1~2条特异性条带;有3个引物(OPK-08、OPN-11和OPN-18)在IRBB4中各扩增出了2条特异性条带;有3个引物(OPL-04、OPN-10和OPO-18)在IRBB5中各扩增出1~2条特异性条带;在轮回亲本中未见有上述类似的片段被扩增,多次重复均获得较一致的结果。因此,推测这些特异性条带(DNA片段)分别与水稻抗白叶枯病抗性基因Xa-3、Xa-4和xa-5相关。
【Abstract】 Bacterial leafblight,caused by Xanthomonas oryzae pv.oryzae,is one of the main diseases of rice.To controll the disease,breeding and cultivation of resistant cultivars is the most economical and effective way.For identification of molecular markers linked to the resistance genes Xa3,Xa4 and xa5,three nearisogenic lines (NILs) IRBB3,IRBB4 and IRBB5,and a recurrent cultivar of IR24 were screened by RAPD.Genomic DNAs extracted from the rice seedlings of the three NILs were analyzed by RAPD using 120 different 10mer random primers.Polymorphic fragments were detected in the three NILs with 8 of the 120 primers.1 to 2 specific DNA fragments were amplified from IRBB3 which contains Xa3 with primers OPK11 and OPN04.2 specific DNA fragments were amplified from IRBB4 which contains Xa4 with primers OPK08,OPN11 and OPN18 respectively.1 to 2 specific DNA fragments were amplified from IRBB5 which contains xa5 with primers OPL04,OPN10 and OPO18.All these polymorphic DNA fragments were not detected in the recurrent parent IR24.Each experiment was repeated several times and the results were the same.So it is inferred that these polymorphic DNA fragments are probably linked to Xa3,Xa4 or xa5.The more detailed analysis is going on to prove the reliability of linkage.
