含人铜锌SOD基因逆转录病毒重组体的构建
CONSTRUCTION OF A RETROVIRAL RECOMBINANT CONTAINING HUMAN CuZn-SOD GENE
【摘要】 目的:构建含人铜锌SOD基因逆转录病毒重组体。方法:通过两步克隆将编码人铜锌超氧化物歧化酶(hSOD1)基因片断从表达载体Bluescriptll中切下,定向插入逆转录病毒载体XM-6上,以限制性内切酶切分析法判定插入是否正确。结果:限制性内切酶谱分析与理论计算相符。结论:含人铜锌SOD基因逆转录病毒重组体构建成功,为今后进行心肌保护、抗动脉粥样硬化及抗衰老的基因治疗等开辟了广阔的前景。
【Abstract】 Objective: To construct a retroviral recombinant containing human CuZn-SOD gene. Methods:With two steps of molecular clonging, a fragment coding human copper/zinc superoxide dismutase(hSOD1) gene was digested from recombinant BluescriptII/SOD(+), and inserted into the vetroviralvector XM-6. The positive clonies were analysized by using restriction mapping. Results: The structureis proved right by restriction mapping. Conclusion: A retroviral recombinant containing hSOD1 genewas obtained. Its expression and gene therapy used for ischemia, arteriosclerosis and aging are underway.
