【摘要】 为了探明抗骨巨细胞瘤（ＧＣＴ）肿瘤细胞成分的单克隆抗体ＧＣＦ－５相应抗原的性质，并为研究ＧＣＴ肿瘤抗原打下基础。本研究采用高浓度ＳＤＳ提取细胞膜蛋白。用ＳＤＳ－ＰＡＧＥ，ＷｅｓｔｅｒｎＢｌｏｔｉｎｇ分析了抗原蛋白的分子量。用蛋白质化学染色技术对ＧＣＦ－５抗原蛋白进行了糖、脂分类染色。结果显示主要的ＧＣＦ－５抗原为分子量１３．５ＫＤ和１７．４ＫＤ的含糖量极少的糖蛋白更多还原

【Abstract】 nordertounderstandthetumorantigen(s)ofgiantceltumorofbone(GCT),monoclonalanti-bodyGCF-5developedinthislaboratorywasemployed.GCF-5reactsedspecificalyagainstthetumorcelassociatedantigensofGCT.Themembraneproteinsoftumorcelwereextractedbyhighconcentra-tionofSDS,folowedbyaseriesoftreatmentprocesses,includingSDS-PAGE,westernblotingandsomestainingmethods,suchasPeriodic-SchifforglycoproteinandoilredOforlipoprotein.Themolec-ularweightandchemicalcharacteristicsofGCF-5antigenswereanalysed.Thepreliminaryresultsindi-catedthatthemainGCF-5antigensweretwoglycoproteiniscontainingfewsaccharidechains,oneofwhichwas13.5KDandtheother17.4KD.更多还原
