硒和维生素E对培养心肌细胞脂质过氧化损伤的协同保护效应

【摘要】 应用氢过氧化枯烯导致培养大鼠心肌细胞脂质过氧化损伤,观察硒和维生素 E 的添加对其影响。结果表明:单纯加硒或维生素 E,均能明显减轻其脂质过氧化物蓄积、肌酸激酶漏出及细胞膜流动性降低,但对~3H—TdR 掺入无显著作用;单纯加维生素 E 还能减轻其肌球蛋白轻链肌酸激酶和头部ATP 酶活性的降低,而硒无此效应;联合添加硒和维生素 E 时,显示出比单纯加硒或维生素 E 更为显著的保护作用;此外,尚能对抗~3H—TdR 掺入的降低,体现出两者之间的协同保护效应。

【Abstract】 Lipid peroxidation was induced in cultured neonatal rat cardiac cells with cumene hydroperoxide( 0.lmmol/L)and the protective effects of selenium or/and vitamin E on the myoeytes injured by lipid peroxidation were investigated.The results showed;addition of selenium(0.1μg/ml)or vitamin E (200lzg/ml α-tocopheral)alone could abate the increase of lipid peroxide in the cells(p<0.05),the leakage of cntraeellular ereatine phosphokinase(p<0.05)and the decrease of the cell membrane fluidity (p<0.01)induced by lipid peroxidation,but had only slight effect on the incorporation of ~3H-TdR into DNA(p<0.05).The effects of vitamin E were better than those of selenium,otherwise,vitamin E could relieve the decrease of the activities of creatine phosphokinase in myosin light chain and ATP en- zyme in myosin head(p<0.01).The effects of administration of both seleium and vitamin E were more powerful than those of selenium or vitamin E alone.Both selenium and vitamin E treated together could relieve the decrease of the incorporation of ~3H-TdR into DNA induced by lipid peroxidation,which was not influenced by selenium or vitamin E alone,that is,the addition of both seleniun and vitamin E showed very strong synergism.These findings provided laboratory evidences for the hypothesis that the combined deficiency of selenium and vitamin E is the main cause of Keshan disease.
