高效毛细管电泳的离柱安培法检测

【摘要】 设计了一种新型的毛细管电泳离柱安培法检测装置,无需截断毛细管,直接使用氢氟酸蚀刻毛细管管壁制作导电接口,可有效地耦合毛细管电泳分离和安培法检测,隔离高压电场对安培法检测的干扰,实现多组分的高效分离和检测。所设计的系统性能优良,导电接口制作简便,耐用性好,无附加体积。

【Abstract】 in this paper, a simple system for interfacing off-column amperometric detection with capillary electrophoresis is demonstrated in 50μm i. d. capillary. HF acid was used to etch the exposed section of capillary column to construct the joint without fracturing the capillary column. When the thickness of capillary wall is less than 20μm after etching, the exposed section will become a porous glass membrane which allows only small buffer ions to pass through it and can be used to form a conductive joint. The amperometric detector was effectively isolated from the high voltage electric field by coupling with this etched joint. Being compared with the porous joints reported by other groups , the fabrication of the etched joint is very simple and inexpensive without the need of porous tubing or porous membrane. Application of the off-column amperometric detection for electrophoretic separation of phenols was demonstrated. High resolution and low detection limits were obtained with this system.
