【摘要】 以２３只家兔分成生理盐水对照组及１０、３０、６０和１００ｋＵ／ｋｇ体重不同剂量组，静脉注射国产高分子量尿激酶，观察给药前后不同时间兔血浆尿型纤溶酶原活化素水平的动态变化，测得其血浆半衰期为３５ｍｉｎ。在２９例急性心肌梗塞患者以８００～１１００ｋＵ的ＵＫ作溶栓治疗时，除测定治疗前后血浆尿型纤溶酶原活化素外，还测定了血浆ｔ—ＰＡ抗原、ＰＡ和相应抑制物ＰＡＩ活性，分析了以大量ＵＫ作溶栓治疗的有效性和相对安全性。更多还原

【Abstract】 In this report the domestics of high molecular weight urokinase(UK)was intravenously injected with different doses of 10000, 30000,60000 and 100000 units per kilogram body weight in 10 ml of physiological saline(PS) into total of 17 rabbits over 20 min through the ear vein.Another 6 animals were given PS as controls.The dynamic variations in plasma urokinase-type plasminogen activator(u-PA-antigen levels were estimated by measuring the u-PA concentra tions in plasma samples obtained before administration, in 1 min after injection,and at various time points during the period of 4h after injection in all dose groups. The half-life of the u-PA in the rabbit blood circulation was calculated as 35min. When 19 cases of acute myocardial infarction (AMI)took the thrombolytic therapy with a bolus dose of 800 000￣1100 000 units UK in lh period for the different individuals, the bold samples were obtained before administration and at various time points during 24h period after the end of injection for measuring u-PA and t-PA antigens as well as PA and PAl activities.According the results the mechanism of variations in these thrombolytic indexes for UK injection and the validity as well as the relative safety of using UK as a kind of thrombolytic drug were discussed.更多还原
