淋巴因子对PMA活化的扁桃体B淋巴细胞CD23分子表达的影响

【摘要】 研究了人淋巴因子rhIL-4、rhIL-2、rhIFN-γ对PMA活化的扁桃体B淋巴细胞CD23分子表达的调节作用。实验结果表明:1.rhIL-4能够显著增加PMA作用下的B细胞CD23表达,呈剂量依赖性反应;rhIFN-γ对活化后B细胞CD23表达呈明显抑制作用;rhIL-2对CD23表达的抑制作用微弱。2.在PMA单独刺激下B淋巴细胞CD23的表达在第3天达高峰,而PMA与IL-4共同作用下IL-4能够持续促进PMA作用下的扁桃体B细胞CD23的高表达,在作用的第5天CD23阳性率可高达75.4%。由此可见B细胞CD23分子的表达受到T细胞源性淋巴因子的调控,这种调控对于免疫应答的调节可能具有重要意义。此外对B淋巴细胞经PMA及淋巴因子活化后,反映细胞分裂状态的 ̄3H-TdR、 ̄3H-UR掺入cpm值与CD23表达的相关性进行探索。发现两者无明显相关性。

【Abstract】 The effects of T cell derived lymphokine rhIL-4, rhIL-2 and rhIFN-γ on CD23 expressionon PMA activated B cell were analysed. The results showed that rhIL-4 could obviously elevatedCD23 expression. The optimum conceneration of rhIL-4 was at 50~100U/ml.Unlike rhIL-4,rhIFN-γ significantly reduced CD23 expression. rhIL-2 could also suppress the CD23 expres-sion,but the effect was much weaker compare with rhIFN-γ. The time effect on CD23 expres-sion on B cell stimulated bv PMA or PMA+IL-4 were also analysed. The results show thatCD23 expression on B cell actived by PMA reached the maximum(44.4%)on the third day.otherwise,CD23 express on B cells actived by PMA+rhIL-4 steadily increased during 5 daysobservation and the percentage of CD23 positive cells fifth day reached 75.4%。 In addition,weobserved the relatiohship between the percentage of CD23 ̄(+) B cells and ̄3H-rdR &  ̄3H-UR incor-poration in activated B cells, The results showed that the change of incorporation value( ̄3H-TdR &  ̄3H-UR) did not cause responsably the change of CD23 expression on actived B cell.
