周期性与连续性给予羟乙膦酸钠对去势大鼠骨质疏松症治疗作用的比较

【摘要】 用不脱钙骨切片的骨组织形态计量学方法比较周期性口服、周期性腹腔注射和连续性口服羟乙膦酸钠(HEBP)对去势雌性大鼠骨质疏松的治疗作用。剂量均为每日36mg/kg。周期为连续5日,停药25日后再重复循环,共90日。结果表明周期性口服HEBP的大鼠较单纯去势大鼠的骨转换率降低,胫骨骨小梁骨量显著增加,从而可减缓骨质疏松程度;连续性口服HEBP的大鼠虽然骨小梁骨量也显著增加,但骨转换率不仅较单纯去势大鼠,也较正常大鼠显著降低,骨小梁质量欠佳,骨重建受损;周期性腹腔注射HEBP的大鼠骺板显著增厚,骨小梁类骨质显著增加,软骨基质和骨矿化均受损。此为临床应用HEBP防治骨质疏松提供可借鉴的资料。

【Abstract】 To compare the effect of intermittent cyclical with continous administration of etidronate disodium(HEBP), 57 female Wister rats. weighing an average of 2659, were randomly divided into 5groups. Group lwas bilateral sham-ovariectomized; group 2 was bilateral ovariectomized(OVX); group 3 was OVX and then the intermittent cyclical HEBP was given orally; group 4 was OVX and then the continous HEBP was given orally; group 5 was OVX and the intermittent cyclical HEBP was given by intraperitoneal injection. HEBP was given during fasting for 5 days followed by a rest period of 25 days and the intermittent cycles were continued for 3 and 90 days, respectively. The proximal tibial metaphses were processed undecalcified for quatitative bone histomorphometry and histological observation.The results are as follows. Group 3 presents more trebecular bone volume than that of group 2,and the elevated bone turnover hy OVX is reduced properly indicating the scheme of group 3 could retard the degree of osteoporosis. Group 4 presents more trabecular bone volume, but its bone turnover is more lower than that of not only group 2 and 3 but also group 1, the appearance and arrangement of trabeculae are irregular, meaning the disturbance of bone remodeling. Group 5 exhibits the accumulation of cartilage metrix in epiphyseal plate resulting in abnormally thickened and the increase of osteoid in metaphyses indicating the highly inhibition of mineralization of bone.
