小儿再生障碍性贫血骨髓细胞核仁形成区蛋白定量研究

【摘要】 目的探讨小儿再生障碍性贫血骨髓细胞的增殖活性。方法收集正常骨髓8例,再生障碍性贫血19例,每例取2~3张骨髓涂片以胶质银技术进行AgNOR染色。油镜下依细胞系列每类细胞计数8~100个细胞核内的核仁形成区蛋白(AgNOR)的颗粒数,求均值,以t检验进行统计学处理。结果急慢性再障骨髓细胞AgNOR均值显著低于正常骨髓组,依次以巨核、早中幼红、早中幼粒细胞下降最明显(P<0.001,P<0.05)。急慢性再障之间AgNOR均值也有显著性差异。单核及淋巴细胞无改变。结论AgNOR技术对小儿再生障碍性贫血的诊断、疗效判定及骨髓细胞增殖障碍机制的探讨有一定价值。

【Abstract】 Objectve To sesrch the proliferative activityof bone marrow cells in children with aplasticanemia.Methods Eight normal children and 19 casesof aplastic anemia were included in this study. Ineach case, 2~3 bone marrow snears were stainedby the colloid silver technique. The AgNOR dotswithin nucleus were enumrated with an optical microscope under an oil immersion lens. The numberof cells examined belonging to a particular typeranged from 8 to 100 in each case. Calculating themean value, t test was used for statistics.Results The AgNOR mean value of acute andchronic aplastic anemia were significantly lowerthan those of control group. The decline ofAgNOR mean value was the most prominent inturn in megakaryocyte, prorubricyte/polychromatic normoblast and promyelocyte/myelocyte (P< 0. 001, P< 0. 05, respectively).There was also a significant difference betweenacute and chronic aplastic anemia.No change wasfound in monocyte, lymphocytes, ect.Conclusion AgNOR technique possess certainpractical value for diagnosis of children’s aplasticanemia, judging theraputic effect and investigating the mechanism of marrow proliferative disturbance.
