三种轮状病毒ELISA试剂的比较研究

【摘要】 作者用自制轮状病毒单克隆抗体ELISA试剂检测细胞培养和88份粪样中轮状病毒,与丹麦Dako公司和上海市卫生防疫站制备的轮状病毒ELISA试剂进行了比较。结果自制试剂的细胞培养中轮状病毒最低抗原检出量是0.03TCID50;对低轮状病毒核酸含量粪样的阳性检出率为95.7%,均高于另二种试剂;检测88份粪样中轮状病毒,自制试剂的灵敏度、特异度及Youden指数分别为98.3%、90%及0.88,与另二种试剂无显著性差别(P>0.05)。此外,自制试剂的阳性标本与阴性标本OD值均数之比约为另二种试剂的2.2倍。实验结果提示,本研究自制轮状病毒ELISA试剂检测轮状病毒的敏感性和特异性较高,可以替代同类轮状病毒多克隆抗体ELISA试剂,且易于目测判断结果,适于基层临床使用。

【Abstract】 In this study the authors tested rotavirus in cell culture and 88 fecal specimens by using a self-made monoclonal ELISA diagnostic kit, and comparison was made with those by DAKO ELISA kit and the kit prepared by Shanghai Antiepidemic Station. The minimal amount of rotavirus antigen detected from cell culture with the self-made kit was 0.03 TCID50 and the detection rate of antigen from the fecal specimens containing only small quantities of viral RNA was 97.5%, both figures being higher than those by the other two kits. The sensitivity, specificity and Youden index of the self-made kit were 98.3%, 90.0% and 0.88 respectively , without significant difference from those of the other two kits. Futhermore, the ratio of mean OD values of positive to negative samples was 2.2 times higher than those of the two others. It suggested that our new monoclonal antibody rotavirus diagnostic kit might replace the commercial kits in clinical diagnosis of rotavirus infections and it would be more suitable for grass-roots health-care institutions.
