米索前列醇对大鼠的胃细胞保护及抗分泌作用
GASTRIC CYTOPROTECTIVE AND ANTISECRETORY EFFECTS OF MISOPROSTOL IN RATS
【摘要】 大鼠禁食后纯酒精灌胃可产生广泛的胃粘膜坏死,预先经口给前列腺素E1衍生物,米索前列醇(Miso)剂量分别为10,30,100μg/kg可预防胃粘膜坏死,其作用有剂量依赖性。应用禁食大鼠幽门结扎试验,Miso1mg/kg可明显降低胃酸浓度及酸分泌量,但对胃液分泌量无影响。结果提示Miso具有抗胃酸分泌及胃细胞保护作用,该化合物可为临床治疗溃疡病提供新的有效方法。
【Abstract】 Oral adminmistration of absolute ethanol to fasting rats produced extensive necrosis of the gastric mucosa. Pretreatment with oral Misoprostol(Miso),a synthetic derivative of prostaglandin E1,at doses of 10, 30 and 100μg/kg prevented sach necrosis, and the effects were dose-dependent. After pyloric ligation in fasting rats, intragastric administration of Miso 1 mg/kg produced significant inhibition of total gastric acid output and acidity but little change in volume of gastric fluids.The results suggest that Miso is an agent with selective gastric antiseoretory and cytoprotective properties that should provide a new therapeutic approach for treatment of peptic ulcer diseases.
