红豆草下胚轴及叶片悬浮细胞系的建立与植株再生

【摘要】 以红豆草栽培品种埃斯基的下胚轴和叶片为外植体,进行初始愈伤组织的诱导,研究了初始愈伤组织的继代、改造和获得松脆愈伤组织的主要影响因素,并继而建立了下胚轴和叶片两个良好、稳定且具分化潜力的悬浮细胞系,两系再生植株的分化率分别为13%和57.1%。此外,还探讨了影响叶片悬浮细胞系分化的主要因素,结果表明:采用二步分化法,在将液体悬浮培养转入固体分化培养时,必须采用对数生长期的悬浮细胞。并在分化培养基中添加较高浓度的细胞分裂素,可获得较高频率的再生植株。

【Abstract】 The calli were inflated from hypocotyls and leaflets discs of Eski Sainfoin (Oncbrychis viciae folia Scop. ) which was screened from six cultivars through repeated subculture and selection yellowish, fast-growing , friable calli (Type II) were obtained. Stable hypocotyl and leaflet disc suspension cell lines, which were mainly comprised of aggregates of small,round > densely cytopasmic and starch - containing cells were established with the type I calli. Non -rooted plantlets were with 13% of quasi - differentiation frequency regenerated from hypocotyl cell suspension line after a period of transition culture. The green plantles were frequently regenerated from leaflet disc suspension cell line, the quasi -difterentiation frequency was as high as 57.1%.Futhermore, the main factors which influenced friable calli were studied, and all kinds of calli were compared on the morphlogy and cytology. We also studied the factors which affected the differentiation frequency of suspension cell line. The results showed that; (1) Shoots were induced effectively with highter concentration of cytokinin (6 mg. L-1 KT). (2) The leaflet disc suspension cell line of logarithmic growth phase (2~ 8) days was apt to regenerate, the highest quasi - differentiation frequency could reach 57. 1%.
