斑点杂交试验检测血清中HBV—DNA的临床应用及其意义

【摘要】 本文用地高辛探针和生物素探针做斑点杂交平行检测了160例乙型肝炎患者,其符合率为96.25%;HBeAg阳性者HBV-DNA检出率为88.41%;而HBeAg及抗HBe均阴性者的阴性率为95.95%。用地高辛探针对21例正常人,28例非乙肝患者中均未检出HBV DNA。实验表明,地高辛探针做斑点杂交具有较好的敏感性特导性和实用性。

【Abstract】 This paper reports that spot hybridization test with a digoxintim labelled probe and a biotin labelled probe was used to detect serum samples from hepatitis Bvirus ( HBV ) -DNA of 160 patients. The result showed that the postive and negative accord-rates between digoxinum labelled probe and biotin labelled probe were 96.25%. Detectable rate of HBV-DNA was 88.41% in HBeAg patients of postive, negative rate of negative patients in HBeAg and anti-HBe were 95.95%. The hybridization procedure using the digoxinum labelled probe was specific as shown by cos-istently negative results in 48 patients without markers of HBV infection. These results indicate that the spot hybridization technique using digoxinum labelled probe is specific, sensitive, reproducible and convenient.
