表皮生长因子受体在小鼠睾丸内免疫组织化学定位
Immunohistochemical Localization of the Epidermal Growth Factor Receptor in Mouse Testes
【摘要】 采用生物素—抗生物素蛋白DCS体系(biotin-avidin DCS system)间接免疫荧光新技术,以176小鼠抗人A431细胞的表皮生长因子受体McAb进行表皮生长因子受体在小鼠睾丸内免疫组织化学定位的研究。在体实验结果显示,小鼠睾丸曲细精管的精原细胞和支持细胞内可见斑点状荧光,其它各级生精细胞均未见荧光。离体实验结果显示,体外培养的小鼠支持细胞内有相当强的荧光。证明精原细胞和支持细胞是表皮生长因子的靶细胞。
【Abstract】 Immunohistochemical localization of the epidermal growth factor (EGF) receptor in mouse testes was studied. The biotin-avidin DCS system indirect immunofluorescent technique was used with mouse anti-human A431 cell EGF receptor monoclonal antibody 176 in vivo. Punctate fluorescence was detected in the cytoplasm of spermatogonia and Sertoli cells in the seminiferous tubules, while no fluorescence was observed in other seminiferous epithelial cells. Strong fluorescence was seen in Sertoli cells in vitro. These results suggest that spermatogonia and Sertoli cells are the target cells of epidermal growth factor.
