【Abstract】 The polymerase chain reaction(PCR) was used to amplify insulin gene fragment (1128bp) in human genomic DNA. The amplified product was directly cloned to MI3 vector, and DNA sequencing was carried out by Sanger dideoxy-mediated chain termination method. This is a simple and effective method for identifying mutations in the coding region of prepeptide, A chain, B chain and C peptide of insulin gene
