水稻线粒体线性大分子DNA的电泳图谱分析

【摘要】 采用脉冲电泳技术分离琼脂糖凝胶薄片包埋的水稻线粒体DNA(mtDN-A),观察到5个主要的DNA电泳区带,其大小分别为20~30kb,60~200kb,1600kb,2000kb和大于2200kb.水稻mtDNA分子的最小长度约300kb.未观察到大小为300kb的水稻主环mtDNA分子.稀有切点限制酶SfiI酶切水稻mtDNA可产生12~200kb的片段,其中某些片段来源于大分子mtDNA.本文对脉冲电泳分离水稻大分子mtDNA的条件与效果、制备方法对线粒体DNA的构型影响以及线性大分子mtDNA的产生原因进行了分析与讨论.

【Abstract】 The genome organization of rice mitochondria was released by pulsed-field gel electrophoresis. The gel patterns showed five different DNA bands of rice mtDNAs. On the basis of data for DNA size standard, these five DNA bands located at 20~30 kb, 60~200 kb, 1600 kb, 2000 kb and more than 2200 kb, respectively. The minimum complexity of the genome was estimated to be 300 kb. No master circular molecule of 300 kb was found in rice mitochondrial genome. Digestion with restriction enzyme results in 12~200 kb fragments, some of which were produced from long linear mtDNAs.
