环磷酰胺诱发小鼠睾丸生殖细胞染色体畸变的有关处理因素研究

【摘要】 作者对环磷酰胺诱发小鼠睾丸生殖细胞染色体畸变的剂量、给药途径和处死时间等实验因素进行了对比研究。结果表明:睾丸病理切片观察见曲细精管内生殖细胞数量减少;各实验组精原细胞和初级精母细胞染色体畸变率均较对照组明显升高;两种给药途径相比和不同剂量之间相比(除25mg/kg外)染色体畸变率均未见明显差异。精原细胞畸变率6天处死组高于13天处死组,而初级精母细胞则正好相反。并出现了三价体和四价体,由此,作者认为,环磷酰胺50mg/kg×5,第6天和13天分批处死动物制片是较理想的阳性对照动物模型。

【Abstract】 A comparative study Of the factors(dose, route of exposure to tested chemicals, times ofthe animal sacrificed) in the test of chromosomal aberration in mouse germ cells induced bycyclophosphamide (CP) was conducted.It was found that morphological observations showed the number of germ cell decreasedafter exposure to CP(100mg/ kg×5), and the rate of chromosomal aberration rosesignificantly in all the test groups. However, there were no significant differences between thedifferent routes of exposure( ip and po) and the different dose groups except for the25mg/kg dose group on the 6th day, the rate of spermatogonium chromosomal aberrationwas higher than that on the 13th day, but in primary spermatocytes, not only more fragments were seen, but also the trivalent and quadrivalent were seen on the 13th day.It is therefore suggested to give CP at the dose of 50rag / kg×5 and sacrifice the animalson the 6th and 13th days when CP is used as a positive control agent in the test of mouse germcell chromosomal aberration.
