旱芹人工种子人工胚乳成分的筛选

【摘要】 试验对人工胚乳中一种大量元素和一种微量元素的含量分别进行了筛选。改良了大量元素的MS-2人工胚乳,可使人工种子的转株率提高到92%,微量元素的试验显示MS-2为最优。对人工胚乳中糖成分及浓度也进行了筛选。在葡萄糖、蔗糖、麦芽糖三种寡糖中,麦芽糖6%浓度时人工种子转株率最高,为94%;蔗糖略低;葡萄糖最低。糖浓度为3%、1%时转株率下降较多。旱芹胚状体在马铃薯提取液或可溶性玉米淀粉为唯一碳源的人工胚乳中不能成苗。

【Abstract】 Selections of concentrations of a macronutient and a trace element in some artificial endesperms show that MS-2c is the best one.Carbonhydrates for the artificial endosperm have also been selected. Among glucose, sucrose and maltose, the highest pfr. 94% emerged when 6% maltose was used for endosperm of the artificial seeds. These carbonhydrates could be arranged in order of pfr and from high to low : 6% maltose, 6 sucrose, 6% glucose; and while sugar concentrations were reduced to 3% or 1%, the pfr . decreased greatly. Another result showed that embryoid could not form seedling when it was coated in endosperm using tomato extract or maize starch as the sole carbon source.
