卵巢癌腹水免疫复合物解离抗原制备单克隆抗体
PREPARATION OF MABS WITH ANTIGENS(OC41) DISSOCIATED FROM IMMUNE COMPLEXES EXTRACTED FROM OVARIAN CARCINOMA ASCITES
【摘要】 本文从卵巢上皮癌腹水免疫复合物中,解离出患者的自身抗原(OC41)免疫小鼠,取其脾细胞用于融合,经ELISA、PAP及流式细胞计等方法筛选和鉴定,获得2株杂交瘤细胞系,分别命名为COC41D和COC41E,2种细胞诱生小鼠腹水,ELISA测定抗体滴度达2×105和1×105。免疫组化分析表明,COC41D和COC41E与卵巢上皮癌具有较高的特异性,阳性率分特为70.2%和52.5%。本研究表明,应用腹水免疫复合物解离抗原有可能获得特异性较高的单克隆抗体。
【Abstract】 We immunized BALB/c mice with antigens (OC41) dissociated from immune complexes (ICs) ofepithelial ovarian cancer ascites. The spleen cells were used for fusion with Sp 2/0-Ag 14 murinemyeloma call line. By cloning and screening, two hybridoma cell lines were obtained from 10 fusions.named as COC41 D and COC41 E respectively. The titers of the 2 MAbs in the ascites of BALB/cmice were 2×105 and 1×105. Immunohistological analysis showed that COC41 D and COC41 E werepositively stained in paraffin embeded tissue sections of ovarian carcinoma as 70.2% and 52.8%tespectively, however, no cross-reactivity with varous kinds of benign and normal human tissuesections was tested. The use of antigens dissociated from autologous ICs as immunogens for preparinghigh specific anti-tumor MAbs may provide a valuable means in the stud of tumor antigens.
