人ω1型干扰素基因在大肠杆菌中的表达

【摘要】 <正> 人ω型干扰素(huIFN-ω)是继α、β、γ后被发现的第4种干扰素型别。IFN-ω除具有与IFN-α相似的抗病毒功能外,在结构上还与动物滋养层干扰素高度相似,因此它既是人体滋养层蛋白启蒙研究的工具,又可能为人类生殖生物学研究提供直接信息。本文作者曾采用聚合酶链反应(PCR)技术首先在国内分离和克隆了IFN-ω1结构基因,这一初始克隆包括完整的成熟多肽及部分信号肽编码区。在此基础上,本文又采用PCR技术将
【Abstract】 Human IFN-ω1 which had recently been identified and isolated in our laboratory, was engineered via polymerase chain reaction(PCR)to a form that could be expressed as a nonfusion protein in E.coli cells. Expression with high efficiency was obtained and the recombinant IFN-ωl possessed antiviral activity as high as 6.50×107U/L. Considering the high structural similarity between IFN-ωl and trophoblast protein,further studies dealing with the significance of IFN-ωl in human reproductive biology is being taken in our laboratory.
