【摘要】 本文报告在 Mg⁺⁺存在下,将一种能特异地连接凝血因子Ⅸ的单克隆抗体偶联至 Affi-Gell5凝胶上,组成亲和层析柱,新鲜血浆通过该柱时, 唯独因子Ⅸ被吸附,流出液即为成品,以此制备缺因子Ⅸ基质血浆。成品内残余的因子Ⅸ极少（<1%）,而其他凝血因子的活性基本末变。分别用本成品和重症乙型血友病病人血浆测定因子Ⅸ活性,再将两者的结果进行比较,相关系数 r=0.93（n=26）,表明本成品可作为重症乙型血友病病人血浆之代用品。本法制备缺因子Ⅸ基质血浆快速、简便,仅需一步即可完成。更多还原

【Abstract】 A monoclonal antibody specific for the magnesium-stabilized conformation of Factor Ⅸ(FIX)was coupled to Affi -Gel 15 to constitute an affinity chro- matographic column.When the fresh plasma passes through this column in the presence of Mg,its FIX was adsorbed exclusively,and the outflow is the finished product,a FIX-defi- cient plasma(FIX DP),with the residual FIX being very few(<1%)and the amount and activity of other co- agulation factors being basically un- changed.The result in the determination of activity of FIX using this product was compared with that using the plasma in patient with severe hemo- philia B,with equivalence coefficient r=0.93(n=26),indicating that this product can be used as substitute for plasma in patients with severe hemo- philia B.This method of preparation of FIX DP is rapid and simple,and can be performed for only one step.更多还原
