【摘要】 本实验主要根据树突状细胞(DC)的半粘附和缺乏Fc受体的特性,进行DC的制备和鉴定。实验证明,脾脏单个核细胞径2h培养后,洗去非粘附细胞,继续培养16h,收集脱落DC后,再经Ig包被板的重复贴壁纯化,除去FcR~+的巨噬细胞,即可获得较纯化的DC。DC在混合性淋巴细胞反应中起着重要的辅佐作用。更多还原

【Abstract】 The spleen dendritic cells (DC) of mice can be enriched to about 90% purity by two steps of adherence to cell culture dish, based on its semi adherent activity and lacking of Fc receptor (FcR).The procedure i nvol ves preparat i on of adherent cells by removal of non-adherent cells after 2 hour-culture, followed by anot her 16hour-culture. and then the semi-adherent cells were col1ected. The containing monocytes were removed by panning on plates coated with mouse Ig. The DC thus were prepared and was potent to induce T respondent cell proliferation in MLR as accessory cells.
