大鼠脑皮层微血管的制备及其去甲肾上腺素水平和多巴胺-β-羟化酶活性测定
NORADRENALINE AND DOPAMINE-β-HYDRO-XYLASE IN THE PREPARATION OF CERE-BRAL MICROVESSELS OF RAT
【摘要】 应用牛血清白蛋白(BSA)漂浮及玻璃珠过滤技术制备脑皮层微血管。用放射酶法测定脑微血管内NA水平为3.806±0.350ng/BMV蛋白mg,用分光光度法测定脑微血管内DβH为27.56±2.37活性单位(1活性单位为nm鱆胺/BMV蛋白mg/小时),提示脑微血管具有生物活性并存在肾上腺素能神经的调控。
【Abstract】 Cerebral mierovessels were isolated from rat with albumin floatation and glass bead filtration technique. The purity of the preparation was confirmed by microscopic observation. The preparation was predominantly composed of capillaries and free from neuron and gila. The diameter of microvessels is about 10μm.The noradrenaline content is 3.806±0.350ng/ BMV pr. mg by radioenzymatic assay and dopamine-β-hydroxylase activity is 27.59±2.36 Unit/BMV pr. mg by spectrophotometric assay in the cerebral microvessels of rat. It is suggested that cerebral microvesselsare regulated and modulated by the adrenergic neural system in the brain.
