【摘要】 参考白细胞介素2(IL2)的制备和活性单位的滴定。取伴刀豆蛋白A(Con A)诱生的大鼠脾细胞培养上清液(粗制IL2)为IL2源,诱生时培养液中加入牛血清白蛋白替代牛血清,以排除血清成分的混杂。粗制IL2经Amicon系统加压浓缩和充分透析后,再经SephadexG 200凝胶柱层析纯化,得到2240 IL2活性单位(u)/ml的纯化IL2。纯化IL2经稀释调正至60IL2u/ml即参考IL2。更多还原

【Abstract】 In this study we report the purification of rat interleukin (IL) 2 and preparation of standard IL 2. Supernatants from a 24h incubation culture of Oon A-stimulated rat spleen cells were collected and concentrated by Amicon ultrafiltration. This was then applied to a Sephadex G200 column (2×106 cm) that had been previously calibrated with the following markers: bovine serum albumin (1.112 2×10-22kg), pepsin (5.810 0 × 10-23kg), and cytochrome 0 (1.920 0 × 10-23kg). Active fractions in the IL 2 assay were pooled, concentrated and dialized. The purified IL 2 titer was determined after dilution. 60 units per ml of IL 2 activity will be used as a standard IL 2 in IL 2 assays.更多还原
