微机辅助正交设计对ELISA实验条件的初步探讨

【摘要】 本文用正交设计对用ELISA法检测抗HSV-Ⅰ型IgG抗体过程中四个常见的影响因素(抗原包被温度、洗涤次数、待测血清温育时间、酶结合物温育时间)进行了研究,较简便地确定了用ELISA法检测抗HSV-Ⅰ型IgG抗体的最佳实验条件组合。改进后的方法较原法节省时间近一半,而不影响实验的敏感性和稳定性。运用微机正交设计计算程序,大大提高了工作效率。

【Abstract】 This artical was reported on using latin square design to determine the optimum conditions of ELISA for detecting IgG antibody against HSV-1. the result showed that the bsst conditions were, incubating viral antigen at 20℃ overnight; washing 4 times with PBS-T; adding test serum and incubating 60 minutes at 37℃; washing 4 times; adding HRP-labled anti-human IgG and incubating 60 minutes at 37℃ ;washing 4 times and then adding substrate solution. The process of this test is much more economic than what reported previouslyi for it saves much time though their sensitivity and reliability are the same. Latin square design method reduces the time of the test process and offers much information to be analyzed. Using microcomputer program for latin square design analysis and cacula-tion omits the tedious artificial calculation and makes more efficient work.
