深低温长期保存角膜技术的研究——Capella法及其改良
STUDY OF TECHNIQUE FOR LONG-TERM CRYOPRESERVATION OF CORNEAS: CAPELLA’S METHOD AND ITS MODIFICATION
【摘要】 用自行研制的“简易深低温角膜降温仪”冻存兔及人尸角膜。对冷冻保护剂、降温冷冻及复温方法3项关键技术做了62项比较性研究、简化和改进。发现角膜复温后的处理是影响冻贮效果的重要因素。优选总结了“二步法”冻贮技术,冻贮9~473天的兔及人尸角膜经电镜、酶组织化学及47只兔和20只人尸角膜移植术后观察2~18个月验证,技术简便,保存价格低廉,长期冻贮效果良好。
【Abstract】 In this paper a series of comparative studies of techniques for long-term cryopreservation of corneas by liquid nitrogen were reported. Fr-om March, 1986, 613 rabbit’s and 38 human corneas were cryopreserved,and 62 comparative tests about the three key techniques: cryoprotectivemedium, freezing and thawing methods were systemically studied andmodification was made by using our self-made "simple corneal cryofree-zer". We discovered that the management of post-thawed corneas was avery important factor influencing the effectiveness. We introduced a very simple "two-step" method of freezing technique which was proved tobe very effective, economical and practical. Results of scanning andtransmission electron microscopy, nine kinds of histochemical enzymestainings, together with 47 rabbit’s and 20 human corneal homograftsfrozen 9 to 437 days showed that the "two-step" method was quite eff-ective, and there were no significant differences between it and Capella’s"four-step" method.
