日本血吸虫抗原的纯化及其应用价值的研究——Ⅱ.虫卵抗原五种提取液圆盘电泳的初步比较

【摘要】 用聚丙烯酰胺凝胶圆盘电泳分析血吸虫可溶性虫卵抗原(SEA)五种提取液:H-SEA、U-SEA、H-SEA1、U-C-SEA1、JEU.其显示的电泳区带分别为8、6、1、1、1条。其中 H-SEA1显有1条区带,具有纯度较高,糖蛋白性质的抗原组分。用 ELISA 法检测血吸虫特异性抗体,它有较高的敏感性与特异性.其纯化方法简便有效,操作过程主要是经高速离心后用高氯酸处理,沉淀除去非糖蛋白物质,然后经葡萄糖凝胶柱层析。尿素溶解性虫卯抗原(JEU)亦显示1条电泳区带.
【Abstract】 A comparison of five purified fractions(H-SEA,U-SEA,H-SEA1,U-C-SEA1,JEU)of schislosome soluble egg antigen (SEA)was made by using polyacrylamide gel electrophoresis.The electropho-retie graphs were shown respectively to have 8,6,1,1 and 1 bands.Among the purified fractions,H-SEA1 had higher purity with glycop-rotein in nature.The ELISA method revealed higher sensitivity and specifity in detecting specific schistosome antibody.Its purification was simple and efficient.The main process was the removal of non-glycoprotein substance of the antigen with hyperchloride acid after high-speed centrifigation followed by sephadex column chromatography.In the urea soluble egg antigen (JEU)was also revealed one electroph-oretic band.
